Collection Procedure:

1. Centrifuge the tube within 6 hours of collection, remove the serum/plasma and transfer to a separate screw cap tube and freeze.
2. If the sample is collected in a PPT tube, there is no need to remove the plasma and place in a separate transport tube. It DOES need to be spun within 6 hours of collection and frozen.

Unacceptable Condition:

Causes for rejection include:
1. Insufficient quantity
2. Green top (heparin) tube used
3. Gross hemolysis
4. Specimen not frozen

Method:
Polymerase chain reaction (PCR) amplification and DNA sequencing
Clinical Significance:
This assay identifies HBV genotypes A to H by detection of type-specific sequences in the HBV polymerase gene. Disease progression and response to therapy may be influenced by genotype. This assay also identifies the nucleotide polymorphisms at nt 1764 in the basal core promoter (BCP) and at codon 28 in the precore region.
