Specimen Requirements

Whole Blood for Factor V Leiden and Prothrombin Gene Mutation
Container /Tube:  Lavender top (EDTA) tube or light blue top (sodium citrate) tube
Specimen: 4 mL (minimum volume: 2 mL) of EDTA or sodium citrate whole blood
Transport Temperature: Room temperature
Note: Label the specimen appropriately (whole blood for Factor V Leiden and Prothrombin Gene Mutation).

Serum for Beta-2 Glycoprotein 1 Antibodies, IgG and IgM and Cardiolipin Antibodies, IgG and IgM
Container/Tube: Serum gel tube
Specimen: 2 mL (minimum volume: 1 mL) of serum
Transport Temperature: Refrigerated
Note: Label the specimen appropriately (serum for beta-2 glycoprotein 1 antibodies, IgG and IgM and cardiolipin antibodies, IgG and IgM.

Plasma for all Others
Container/Tube: Light blue top (sodium citrate) tubes
Specimen: 6 mL (minimum volume: 3 mL) of sodium citrate platelet poor plasma
Transport Temperature: Frozen (plasma)

Collection Instructions:
1. Centrifuge specimen in a centrifuge that has been validated to ensure platelet poor plasma.
2. Immediately transfer the plasma to a stoppered, plastic tube and centrifuge again for 10 minutes.
3. Transfer the supernatant plasma into at least 3 screw-capped, plastic vials and send frozen at ≤ -20 °C, if possible.
4. If unable to centrifuge, forward the unprocessed sodium citrate whole blood (minimum volume: 2 mL) to the laboratory promptly at room temperature only.
5. Please complete a "Coagulation Request Form",  list any anticoagulants the patient is receiving, and forward with the specimens.
6. Label the specimens appropriately (citrate plasma).
